Walaupun sudah tidak muda lagi ( 51 Tahun ), Pengalamannya dalam menangani berbagai wabah bioterrorism membuatnya menjadi sosok yang paling tepat untuk menyelidiki rentetan kematian aneh yang terjadi di wilayah Raccoon City. Dengan kemampuan fisik yang semakin matang dan paras yang semakin tampan, kehadiran Leon sangat dinantikan fans dalam menjalankan misinya di Resident Evil Requiem.

Pendatang baru yang memegang peran kunci dalam cerita ini adalah Grace Ashcroft. Ia digambarkan sebagai seorang intelijen analis dari FBI yang punya kemampuan deduksi luar biasa tajam. Ia terus menggali rahasia perusahaan farmasi tersebut hingga akhir hayatnya.
